Single-molecule Visualization of DNA Mismatch Repair

Abstract
DNA mismatch repair (MMR) is a catalytic reaction of a strand-specific degradation and resynthesis dependent on mismatched nucleotides by coordination of sequential signaling pathways. MutS homolog (MSH) binding to the mismatch transmits the mismatch finding ultimately to a distant strand scission site. The resulting downstream signaling leads the excision reaction that proceeds from the strand break to the mismatch either in the 5’ or 3’ direction. However the mechanics of the long distance downstream signaling has been highly controversial for decades and the molecular process of the strand excision has been also puzzling. Single-molecule imaging is capable of elucidating the salient features of a variety of biological processes when individual biomolecules and their complex are followed going through biochemical reactions in real time. We have developed and employed novel single-molecule tools to interrogate the mechanics of the MMR processes [1-5].
